Challenges of RNA Hydrogel

Ribonucleic acid (RNA) is a polymeric molecule that is essential in various biological processes of gene coding, decoding, regulation and expression. RNA can fold itself into complex tertiary structures such as kissing loops and pseudoknots. Although RNA plays an important role in biological processes due to its abundant structural motifs and functional diversity, the practical applications of single RNAs are limited by their inherent thermodynamic and chemical instability, difficulties in predicting structure and stability, and the small scale of production facilities. To achieve self-assembly of RNA into RNA hydrogels without any external support, RNA molecules need to be able to fold themselves into stable tertiary structures and can bind non-covalently to each other through unique intermolecular recognition sequences, thus spanning spatially into networks in 3D polymeric aqueous solutions. Currently, sequence-directed modular design can be used to generate RNA molecules with appropriate sequence motifs to construct RNA hydrogels with desired biological functions and mechanical properties. In addition, chemical modification of RNA monomers is required to make them resistant to ribonucleases for in vivo applications.
