Introduction to Crosslinking Strategies for Hydrogels
Hydrogels are mainly manufactured by crosslinking to create stable polymer networks, of which physical and chemical crosslinking are the two basic strategies. Physically cross-linked hydrogels are hydrogels that are formed through physical interactions such as hydrogen bonding, electrostatic interactions, or van der Waals forces. Typically, physical crosslinking is reversible, and the hydrogels formed by this process can be disrupted by temperature, pH, or ionic strength changes, and are therefore relatively unstable. However, the outstanding advantage of physically cross-linked hydrogels is their biomedical safety and the fact that they are generally more flexible and sensitive to environmental changes. Chemical crosslinks typically involve enzymatically induced crosslink, free radical polymerization-induced crosslink, Schiff base formation, Diels-Alder "click" reaction, oxime formation, and Michael type-addition. It is usually irreversible, and the resulting hydrogels are usually of high mechanical strength and stability, and easily adjustable. Suitable raw materials and crosslinking methods can be selected to produce hydrogels with appropriate properties depending on the specific application goals.
